Test Format: Immunofluorescence Assay (IFA)
Bacterial Ringrot: Bacterial ringrot is a severe disease of potato. Clavibacter michiganensis subsp. sepedonicus (Cms) is the pathogen of potato ringrot and has been reported in all major potato grow areas.
Reactivity: This Immunofluorescence Assay (IFA) reacted with all Cms isolates tested. This IFA procedure requires about 3 hours to perform. Resulted FITC-fluorescenced bacteria on the slides can be visualized by fluorescence microscopy.
Sensitivity: The IFA test is very sensitive and accurate. The potato ringrot bacterium (Cms) can be consistently recognized among other contaminated bacteria and among plant tissues.
Specificity: This test system reacts negatively with Acidovorax avenae subsp. citrulli, Clavibacter flaccumfaciens, Clavibacter michiganensis subsp. insidiosus, C. m. subsp. michiganensis, C. m. subsp. nebraskensis, C. m. subsp. tessellarius, Erwinia carotovora subsp. atroseptica, E. c. subsp. carotovora, E. chrysanthemi, E. stewartii, Pseudomonas syringae pv. phaseolicola, P. s. pv. syringae, Ralstonia solanacearum, Xanthomonas campestris pv. armoraciae, X. c. pv. campestris, X. c. pv. citri, X. c. pv. diffenbachiae, X. c. pv. pelargonii, X. c. pv. phaseoli, and X. c. pv. vesicatoria.
There is no cross reaction with healthy plant tissues. Background is good under fluorescence microscopy views.
Application: The test can be used to detect Cms, pathogen of potato ringrot, in infected host plants as well as in pure bacterial cultures.
